Without knowing all the details, we can only make an estimate. However, for a basic shed that is 12x16 with a relatively lightwight roof, you can probably use 4x4 pressure treated posts to support the structure, spaced 4 feet on center around the perimeter. There are many possible ways to build the shed and the foundation. If you run perimeter joists, such as 2x6 pressure treated, you can run the floor joists the 12 foot direction on 16 inch centers, so long as you have one intermediate girder to support the 2x6 joists at midspan. The girder would run along the 16 foot direction and could probably be a 4x8. You might put a single post in the center of the girder to provide extra support. When I build something I like to build it strong, so this advice may be overkill, but your shed will last a lifetime. Good Luck.
